02 — Mechanism of Action
Snap-8 works through the same fundamental mechanism as argireline but with a broader SNARE protein inhibition that produces more complete reduction in neurotransmitter release.
Like argireline, Snap-8 contains a sequence derived from SNAP-25 that competitively inhibits this SNARE protein. The longer octapeptide chain provides a larger binding footprint with potentially higher affinity than argireline's hexapeptide sequence.
Snap-8's additional sequence extends inhibition to VAMP (vesicle-associated membrane protein) — the third SNARE protein in the fusion complex. By inhibiting both SNAP-25 and VAMP, Snap-8 more completely disrupts the three-way SNARE complex needed for vesicle fusion.
With both key SNARE proteins partially inhibited, acetylcholine release at the neuromuscular junction is reduced more completely than with SNAP-25 inhibition alone. This translates to greater reduction in facial muscle contraction force.
